The Wrong Question
This gets framed as a choice and it usually isn't one: Hong Kong West Kowloon is 47 minutes from Guangzhou South on the fastest trains, at ¥215 second class (MTR's published fare), with immigration for both jurisdictions in the same building. Here is the real answer. Hong Kong is denser, more international, more expensive and more legible to a Western visitor. Guangzhou is cheaper, deeper in Cantonese culture, better value for food at every price point, and far less visited. The practical warning that costs people money: leaving the mainland for Hong Kong uses up a single-entry Chinese visa.

Both, in one trip
Fly into one, train to the other. A common pattern is Hong Kong in, Guangzhou for the middle of the trip, then out from Guangzhou or onward. On the 240-hour transit scheme, mind the routing: Hong Kong can be your entry or your exit, but a Hong Kong–mainland–Hong Kong round trip does not qualify.

Hong Kong and Macau are separate immigration jurisdictions. Crossing into either is an exit from mainland China, whatever the map suggests — and it does not feel like one, because the train takes 47 minutes and both immigration halls sit in the same building at West Kowloon.
If you hold a single-entry Chinese visa, a Hong Kong day trip ends your mainland stay. You cannot return on it. If you entered under China's 30-day unilateral visa-free scheme, none of this applies — each entry is a fresh 30 days and you can cross as often as you like.
It also runs in your favour: Hong Kong West Kowloon is a designated 240-hour visa-free entry port into the mainland. Eligible travellers — including US passport holders — can fly into Hong Kong, take the train north, and enter China visa-free at the station — provided they hold a confirmed onward ticket leaving the mainland within 240 hours for a third country or region other than Hong Kong, such as a flight home from Guangzhou or Shanghai. Going back out to Hong Kong to catch your flight home is a round trip, and the scheme does not cover it.

Should I visit Guangzhou or Hong Kong?
Both, if you can — they are 47 minutes apart on the fastest high-speed trains, at ¥215 second class. If you must choose: Hong Kong is easier, more legible and far more expensive; Guangzhou is cheaper, deeper in Cantonese culture and much better value for food. Guangzhou is also the source city for most of what Hong Kong is famous for culturally.
Do I need a separate visa for Hong Kong from Guangzhou?
Hong Kong has its own entry rules and most Western nationalities enter visa-free. The critical issue is the other direction: if you hold a single-entry Chinese visa, leaving for Hong Kong consumes it and you cannot re-enter the mainland. Under China's 30-day visa-free scheme, each entry is fresh and there is no problem.
Is Guangzhou cheaper than Hong Kong?
Substantially. A good local meal is ¥25 against Hong Kong prices several times that; hotel rooms run ¥180–750 for the bands most travellers use. Guangzhou is the cheapest of China's tier-one cities and Hong Kong is one of the most expensive cities in Asia.
Can I do Guangzhou as a day trip from Hong Kong?
Yes — 47 minutes each way makes it feasible, and guided day tours from Hong Kong exist. But you will spend the day on the old-city cluster and miss the food, which is the actual reason to come. Two nights is the minimum that does the city justice.
Is Guangzhou's food better than Hong Kong's?
Deeper and much better value; Hong Kong's top end is world-class but expensive.
